Can I cut the end off of a speaker cable and wire it up to a speaker to go in a combo amp?

i don't see why not. just make sure you're aware which wire is positive and which is negative, and connect them accordingly.
heavy duty lamp cord works great as well. cheap too.
just make sure it's not shielded guitar cable. You want both wires to be equal sizes, not one a core, the other a shield.

Does it matter what wire I wire to each post? Or does one wire need to correspond with a certain post and a certain spot on the jack?

in a 1/4 inch speaker cable jack, you have the tip and the sleeve. the tip is obviously the pointed tip of the jack, and the sleeve makes up the rest of it. the tip is positive and the sleeve is negative. just be sure to connect them that way to the speaker. and the terminals on the speaker should be labeled or -
