Can I make a calculation of the selling price of an item to arrive at the
cost of the item while using another cell to input a desired mark-up
percentage, without creating a separate cell that contains gross margin?

For example, cell A1 contain the number 100, cell A2 is used to input a
desire mark-up percentage (20 percent). I would like cell A3 to equal 80.Try

=A1*(1-A2)--
